How many generations do I need to have to achieve them all? Is the last default destination is the Tamatomo place?

No destinations unlock as such.
The Friend Plaza opens from the second generation onwards when an adult has gotten all 4 Tamatomo stamps and it's happiness metre is into the last bar.

The Friends Plaza is the only destination to 'unlock'
